Resumo: O câncer representa atualmente um grave problema de saúde pública, e diversas pesquisas buscam desenvolver novas moléculas que possuam propriedades farmacológicas duplas, assegurando melhor eficácia terapêutica e menor efeito colateral. O estudo da relação estrutura-atividade de diferentes protótipos tem revelado importantes moléculas com ação antitumoral. O LQB-278, por exemplo, foi sintetizado a partir da inserção do fragmento terpênico geranila no anel aromático do N-Fenil-t-butilnitrona, e apresentou importante ação antileucêmica in vitro. Este trabalho estudou a interferência de modificações estruturais no terpenila nitrona LQB-278 com relação à ação antineoplásica in vitro, avaliando também o ciclo celular, a morte celular por apoptose e a expressão de p21. A citotoxicidade foi avaliada pela técnica do MTT, sobre linhagens celulares leucêmicas (Jurkat e K562) e tumores sólidos (PC3, A549 e MCF-7); além de células não tumorais (fibroblastos NIH-3T3 e células mononucleares de sangue periférico humano). As células foram cultivadas e tratadas ou não com as substâncias em estudo por 72h. A proliferação celular foi avaliada pelo ensaio de exclusão com azul de Tripan, e o ciclo celular, a morte celular por apoptose e a expressão de p21 foram realizadas por citometria de fluxo em células Jurkat. A leucemia linfocítica Jurkat foi a linhagem celular tumoral que se mostrou mais sensível às moléculas em estudo, com IC50 de 2,6 ± 0,6 µM, 1,4 ± 0,4 µM e 2,8 ± 0,3 µM, para LQB-292, LQB-287 e LQB-461, respectivamente. Tais moléculas mostraram baixa ação citotóxica sobre células não tumorais. Estes LQBs reduziram (p<0,05) a produção in vitro de NO, mas também induziram morte de macrófagos RAW 264.7. Os três LQBs mais ativos reduziram significativamente a proliferação de células Jurkat em todas as concentrações testadas. O LQB-287 aumentou a expressão de p21, sem alterar o ciclo celular ou induzir morte. O LQB-461, nas concentrações de 2 µM e 3 µM, e o LQB-292, na concentração de 1 µM, também não alteraram o ciclo celular ou induziram morte celular. O LQB-461 também mostrou aumento na expressão de p21 nestas concentrações. Por outro lado, o LQB-461 a 5 µM aumentou (p<0.05) o percentual de células na fase sub-G1, sugerindo morte celular, diminuindo G1 e G2/M (9,0± 3,3 %), sem alterar a fase S. O LQB-292 a 3 µM e 5 µM também mostrou aumento (p<0,05) da fase sub-G1, com diminuição nas fases G1, S e G2/M. O ensaio de anexina-V confirmou a indução de apoptose, tanto inicial como tardia, para o LQB-461 a 5 µM e LQB-292 a 3 µM e 5 µM. Podemos concluir que: 1) A ciclização do radical terpenila aumentou a atividade antitumoral, sendo a posição orto e os grupamentos aldeído e nitrona essenciais para sua função. 2) A linhagem Jurkat foi a mais sensível ao tratamento, com baixa toxicidade para células não tumorais. 3) A inibição da produção in vitro de NO, pelos LQBs, não foi muito importante, já que tais moléculas também induziram morte celular; 4) O LQB-287 parece possuir ação citostática, aumentando a expressão de p21; 5) O LQB-461 e o LQB-292 induzem efeitos diferentes em função da concentração, possuindo uma aparente ação citostática, nas menores concentrações, e ação citotóxica, em concentrações maiores, com aumento na expressão de p21 e morte celular por apoptose.
Abstract: Cancer currently represents a serious public health problem, and several studies are searchinh develop new molecules that have double pharmacological properties, ensuring therapeutics efficacy and lower collateral effects. The study of the structure-activity relationship of different molecules has revealed important prototypes with antitumor action. LQB-278, for example, was synthesized from the insertion of the geranyl terpene fragment into the aromatic ring of N-Phenyl-t-butylnitrone, and showed an important in vitro antileukemic action. This work studied the interference in the structural modifications of terpenyl nitrona LQB-278 concerning antineoplastic action in vitro, also evaluating the cell cycle, cell death by apoptosis and expression of p21. Cytotoxicity was measured by MTT assay with leukemic cell lines (Jurkat and K562) and solid tumors (PC3, A549 and MCF-7); In addition to non-tumor cells (NIH-3T3 fibroblasts and human peripheral blood mononuclear cells). The cells were cultured and treated or not with the test substances for 72h. Cell proliferation was assessed by the Tripan blue dye exclusion assay, and the cell cycle, apoptosis and p21 expression were performed by flow cytometry in Jurkat cells. The Jurkat lymphocytic leukemia was the most sensitive tumor cell line to the molecules under study, showing IC50 of 2.6 ± 0,6 μM, 1.4 ± 0,4 μM and 2.8 ± 0,3 μM for LQB-292, LQB-287 and LQB-461, respectively. Such molecules showed low cytotoxic action on non-tumor cells. These LQBs reduced (p <0.05) in vitro NO production, but also induced RAW 264.7 macrophage death. The three most active LQBs significantly reduced the proliferation of Jurkat cells at all concentrations tested. LQB-287 increased p21 expression, without altering the cell cycle or inducing death. LQB-461 at 2 μM and 3 μM, and LQB-292 at 1 μM concentration, also did not alter the cell cycle or induced cell death. LQB-461 also showed increased p21 expression at these concentrations. On the other hand, LQB-461 at 5 μM increased (p <0.05) the percentage of cells in the sub-G1 phase, suggesting cell death, decreasing G1 and G2/M, without altering S phase. The LQB-292 at 3 μM and 5 μM also showed an increase (p <0.05) in the sub-G1 phase, with a decrease in G1, S and G2/M phases. The annexin-V assay confirmed the induction of apoptosis, both early and late, for LQB-461 at 5 μM and LQB-292 at 3 μM and 5 μM. We can conclude that: 1) The cyclization of the terpenyl radical increased the antitumor activity, with the ortho position and the aldehyde and nitrone groups being essential for its function. 2) The Jurkat cell line was the most sensitive to treatment, with low toxicity to non-tumor cells. 3) Inhibition of in vitro NO production by LQBs was not a lot important, since such molecules also induced cell death; 4) LQB-287 seems to have cytostatic action, increasing p21 expression; 5) LQB-461 and LQB-292 induce different effects as a function of their concentration, having an apparent cytostatic action, at lower concentrations, and cytotoxic action, at higher concentrations, promoting of p21 expression and apoptosis cell death.
